<p>Infant mortality is tied to the concentration of lead in the air, according to a new study.</p>



<p>Concentrations of lead in the air remain substantial in developed and developing countries, largely the result of industrial emissions.</p>



<p>Even with the decline in lead in gasoline, developed countries, including the United States, still have substantial air lead emissions and in part as a result,&nbsp;<a href="https://www.futurity.org/kids-lead-exposure-systemic-racism-2969842/">blood lead levels in children</a>&nbsp;are high worldwide.</p>



<p>In a new study, researchers used data on lead emissions in the air to estimate the effects of air lead concentration on infant mortality. The study found a statistically significant relation between air lead concentration and infant mortality.</p>



<p><a href="https://doi.org/10.3386/w33447">The study</a>&nbsp;is a National Bureau of Economic Research Working Paper.</p>



<p>“Although many studies have examined the adverse effects of lead on children’s cognitive and behavioral outcomes, few have analyzed the effect of exposure to lead on infants’ health, so we know little about the extent of harm to infants’ health from airborne lead in settings with modern medical care and at modern exposure levels,” explains Karen Clay, a professor of economics and public policy at Carnegie Mellon University’s Heinz College of Information Systems and Public Policy, who led the study.</p>



<p>Lead is known to cause adverse health effects in humans&nbsp;<a href="https://www.futurity.org/screening-index-maps-chronic-lead-exposure-2469882-2/">across a range of exposures</a>. In this study, researchers used data on lead emissions from the US Toxics Release Inventory (TRI), which was created in 1986 in response to chemical releases in Bhopal in 1984 and in West Virginia in 1985; the TRI covers over 650 chemicals. The causal effect of lead on infant mortality is identified by annual variation in air fugitive lead emissions interacted with wind speed near reporting plants, which together determine local ambient lead concentration.</p>



<p>The researchers also analyzed lead monitor data from the Environmental Protection Agency (EPA)’s Air Quality System, wind data from the National Centers for Environmental Information, and infant health data from the National Vital Statistics system of the National Center for Health Statistics. The study included 127 US counties that have plants with lead emissions within 2 miles of EPA lead monitors and within 10 miles of a wind monitor.</p>



<p>Higher concentrations of lead in the air caused higher levels of infant mortality in infants’ first month and first year, suggesting that both in utero and environmental exposures matter. In addition, higher lead concentration boosted deaths from low birthweight, sudden unexplained infant death, and respiratory and nervous system causes.</p>



<p>“Back-of-the-envelope calculations indicate that declines in fugitive lead emissions prevented 34 to 59 infant deaths per year, generating benefits of $380 million to $670 million annually,” says Edson Severnini, an associate professor of economics at Boston College, who coauthored the study.</p>



<p>“In the United States, industrial firms and the aviation industry still emit hundreds of thousands of pounds of lead into the air,” notes Xiao Wang, assistant professor of economics and trade at Hunan University, who coauthored the study. “Our new estimates can inform investments in reducing air lead emissions and soil cleanups.”</p>

<p>In an extensive and up-to-the-minute review of research in this domain, researchers from the University of California, Irvine illuminate the&nbsp;<a href="https://www.futurity.org/childhood-adversity-death-stress-2425842-2/">profound impacts</a>&nbsp;of these adverse childhood experiences on brain development and introduce new paths for understanding and tackling them.</p>



<p>Their study in&nbsp;<a href="https://doi.org/10.1016/j.neuron.2025.02.016"><em>Neuron</em></a>&nbsp;examines the mechanisms behind the long-term consequences of childhood stress (adversity). Despite extensive research spanning over seven decades, the authors point out that significant questions remain unanswered.</p>



<p>For example, how do adults—from&nbsp;<a href="https://www.futurity.org/adversity-emotional-support-parents-2793102-2/">parents</a>&nbsp;to researchers—fully comprehend what is perceived as stressful by an infant or child? Such conceptual queries, as well as the use of cutting-edge research tools, can provide a road map, guiding experts toward developing innovative methods and providing solutions to this pressing mental health issue.</p>



<p>“Our research suggests that the unpredictability of a child’s early environment may be just as important as more traditionally recognized forms of adversity, such as abuse or neglect,” says lead author Tallie Z. Baram, a professor of pediatrics at the University of California, Irvine and one of the world’s leading researchers in this field.</p>



<p>“Our review has important implications for how we approach early intervention and prevention strategies.”</p>



<p>She and coauthor Matthew Birnie, a UC Irvine postdoctoral scholar, identify several key areas for further investigation:</p>

<li>What does the developing brain perceive as stressful?</li>



<li>Which aspects of stress most significantly influence brain maturation?</li>



<li>Which developmental ages are most vulnerable to adversity?</li>



<li>What are the molecular mediators of stress effects on the brain?</li>



<li>How can transient stressful experiences lead to enduring dysfunction?</li>
</ul>



<p>One notable discovery is a novel form of early-life stress: unpredictable sensory inputs from caregivers and the environment. This factor plays a substantial role in adverse neurodevelopmental outcomes, even after controlling for well-known adverse childhood experiences, which are collectively referred to as ACEs.</p>



<p>The review highlights the limitations of current ACE scoring systems in accurately predicting individual outcomes and underscores the complexity of early-life stress. Emerging factors, such as societal and anthropogenic characteristics like inequality and pollution, are gaining recognition as potential contributors.</p>



<p>Animal models have been instrumental in unraveling the mechanisms underlying brain development effects. Research has revealed that different types of stress can yield distinct outcomes, influenced by the nature and timing of stress, as well as species, strain, and sex variations.</p>



<p>At the molecular level, early-life stress can substantially alter neuronal gene expression through epigenetic mechanisms. These changes may lead to long-term modifications in how the brain responds to subsequent experiences. At the circuit level, early stress can disrupt the maturation of brain networks by interfering with crucial developmental processes, including neuronal oscillations and synaptic pruning.</p>



<p>“We’re gradually comprehending how early-life stress can ‘reprogram’ the brain at multiple levels, from individual molecules to entire neural circuits. This knowledge presents new avenues for targeted interventions,” Baram says.</p>



<p>The review also identifies key molecular mediators of early-life stress effects, including glucocorticoids and neuropeptides like corticotropin-releasing hormones. Ongoing research is uncovering novel roles for these molecules in specific neural circuits affected by early stress.</p>



<p>In light of these findings, the researchers propose redefining early-life stress as “early-life adversity” to better encompass the diverse experiences that can affect brain development, even those not traditionally perceived as stressful.</p>



<p>“This review emphasizes the need for a more comprehensive understanding of early-life adversity,” Baram says. “By focusing on how the developing brain processes and responds to these experiences, we can develop more effective strategies to prevent and mitigate their long-term effects.”</p>



<p>The researchers suggest increased funding for and attention to this critical area of study, highlighting its potential to enhance mental health outcomes and reduce the societal burden of early-life adversity.</p>

<p>MARLTON, N.J., March 12, 2025 /PRNewswire/ &#8212; A growing number of women who received Depo-Provera injections are coming forward with serious health concerns following new research linking the contraceptive drug to an increased risk of meningioma, a type of intracranial tumor. Many individuals who used progestogen-only injections such as Depo-Provera (DMPA, medroxyprogesterone acetate) for extended periods have reported symptoms such as chronic headaches, vision problems, dizziness, memory impairment, and seizures—common neurological symptoms associated with brain masses.</p>



<p>If you or a loved one developed a meningioma after long-term use of Depo-Provera, legal options may be available. However, due to strict statutes of limitations, those affected have only a limited window of time to take action. In some states, the statute of limitations for product liability cases is just one year from the date of injury or discovery. <p>What Is the Connection Between Depo-Provera and Meningiomas?</p>



<p>Depo-Provera is a progestin-based contraceptive shot used by millions worldwide for birth control, as well as for treating endometriosis, menstrual irregularities, and other hormonal conditions. However, a March 2024 study published in the&nbsp;<a href="https://c212.net/c/link/?t=0&amp;l=en&amp;o=4382033-1&amp;h=3427640160&amp;u=https%3A%2F%2Fwww.bmj.com%2Fcontent%2F384%2Fbmj-2023-078078%2Frapid-responses&amp;a=British+Medical+Journal" target="_blank" rel="noreferrer noopener">British Medical Journal</a>&nbsp;uncovered a troubling link. Researchers from France’s National Agency for Medicines and Health Products Safety determined that prolonged use of medroxyprogesterone acetate (DMPA) increases the risk of developing meningiomas by five and a half times.</p>

<p>ViiV Healthcare, the global specialist HIV company majority owned by GSK, with Pfizer and Shionogi as shareholders, today announced positive findings from the company’s EMBRACE phase IIb study. 1 The study found that N6LS (VH3810109 or VH109), given every four months in combination with monthly cabotegravir long-acting (CAB LA), successfully kept viral levels suppressed in adults living with HIV who were already stable on treatment. It was also well tolerated by participants.</p>



<p>These results were presented today at the Conference on Retroviruses and Opportunistic Infections (CROI 2025) in San Francisco, U.S.</p>



<p><em>Kimberly Smith, M.D., MPH, Head of Research &amp; Development at ViiV Healthcare, said</em>:&nbsp;<em></em>“As leaders in long-acting injectable innovation, we are building on the positive patient and physician experience we have with&nbsp;<em>Cabenuva</em>&nbsp;and pioneering the next generation of long-acting treatment options. The EMBRACE study demonstrated that VH109, a CD4-binding broadly neutralising antibody, administered every four months with cabotegravir, achieved high efficacy and was well tolerated through six months. We’re looking forward to continuing the development of VH109 as a component of our future ultra long-acting regimens.”</p>



<p>Results from the EMBRACE study at the six-month primary endpoint showed that 96% of participants receiving VH109 60mg/kg intravenously (IV) and 88% receiving VH109 3000mg subcutaneously (SC) with rHuPH20 maintained HIV-1 RNA levels below 50 copies/mL, compared to 96% in the standard-of-care group. VH109 was administered in both arms every four months, combined with monthly CAB LA. Confirmed virologic failure was observed in two participants from each VH109 group.</p>



<p>Overall, 4% of the IV group and 6% of the SC group had HIV-1 RNA levels of 50 copies/mL or higher, compared to none in the standard-of-care group when measured at month six.</p>



<p>VH109 was generally well tolerated, though infusion site reactions were more frequent with SC administration, occurring in 14% compared to none with IV administration. Adverse events specific to the use of study medication were reported in 64% of the IV group and 65% of the SC group, with 16% of participants in the SC group experiencing grade 3-4 adverse events (erythema). No participants in the IV group experienced a grade 3-4 adverse event.</p>



<p>Based on the favourable results seen in the trial, ViiV Healthcare will be progressing a six-month IV formulation of VH109 in combination with CAB LA for further evaluation in an EMBRACE part two trial.</p>



<p>About&nbsp;<em>Cabenuva</em>&nbsp;(cabotegravir + rilpivirine)&nbsp;<em>Cabenuva</em>&nbsp;is indicated as a complete regimen for the treatment of HIV-1 infection in adults and adolescents 12 years and older and weighing at least 35kg to replace the current antiretroviral regimen in those who are virologically suppressed (HIV-1 RNA &lt;50 c/ml) on a stable antiretroviral regimen with no history of treatment failure and with no known or suspected resistance to either cabotegravir or rilpivirine.</p>

<p>The complete regimen combines the integrase strand transfer inhibitor (INSTI) cabotegravir, developed by ViiV Healthcare, with rilpivirine, a non-nucleoside reverse transcriptase inhibitor (NNRTI) developed by Janssen Sciences Ireland Unlimited Company. Rilpivirine tablets are approved in the U.S. and when used with cabotegravir is indicated for short-term treatment of HIV-1 infection in adults and adolescents 12 years and older and weighing at least 35kg who are virologically suppressed (HIV-1 RNA less than 50 copies/mL) on a stable regimen with no history of treatment failure and with no known or suspected resistance to either cabotegravir or rilpivirine.</p>



<p>INSTIs inhibit HIV replication by preventing the viral DNA from integrating into the genetic material of human immune cells (T-cells). This step is essential in the HIV replication cycle and is also responsible for establishing chronic disease. Rilpivirine is an NNRTI that works by interfering with an enzyme called reverse transcriptase, which stops the virus from multiplying.</p>
